memory. Although the eluates were collected and stored on line with the chromatograph, they were
analyzed later at some convenient time. A diagram of their apparatus is shown in Figure 8.9.

The sample valve (Valco) had an internal loop, 1.9 µl in volume and the column was 17 cm long, 1.1
mm I.D., packed with Rosil C18 reversed phase, particle size 5 µm. The mobile phase was a
methanol/water mixture (either 95:5 or 80:20) used at a flow rate of 20 µl/min. The IR data was
obtained from a Bruker 1FS-85FT-IR spectrometer, equipped with a Bruker A590 FT-IR microscope.
The microscope contained a 16x Cassegrainian lens and a narrow-range mercury-cadmium telluride
detector. The microscope had an adjustable rectangular aperture and normally 128 scans were co-added.
