Analogous sulfonyl chlorides (Fig. 3.6), dabsylCl [31], mansyl-Cl [32], phisyl-Cl [33], BHBTSOCl
[34], also act on primary and secondary amino compounds in a similar manner.
General Derivatization Procedure of Amines with BHBT-SOCI
To 100 µl of a test solution of amines in acetonitrile are added 100 µl of 10 mM triethylamine in
acetonitrile and 100 µl of 10 mM BHBT-SOCI in acetonitrile. The mixture is allowed to stand at room
temperature for about 2-3 min, then a portion (20µl) is injected into RP-HPLC. The fluorescence
intensity is monitored with excitation at 330 nm and emission at 450 nm. The detection limits for
primary and secondary amines are about 3 and 300 fmol, respectively, on column. Fig. 3.7A shows a
typical chromatogram of BHBT derivatives of some amines.
